Jerusalem is a town in Yates County, New York. It is in the Finger Lakes region of New York State. The town sits next to Keuka Lake and forms a peninsula between the two lake portions. The area was first settled in the 18th century by Quakers, who gave the town its name. Jerusalem makes up sixty-five square miles, mostly farmland. There are numerous vineyards throughout the town. Jerusalem consists of several hamlets and one village. The hamlets are Bluff Point, Branchport (formerly "Esperanza"), Friend, Guyanoga, Keuka Park, and Yatesville. The village is named Penn Yan. Penn Yan is split between three towns. Most of the village is in the town of Milo. Smaller portions of the village are found in Benton and Jerusalem., the total town population was estimated at 4,300.

It continues west until it reaches the Hamlet of Jerusalem. There it intersects with CR-29, which runs north and south. The NYS-54A also meets the CR-32 which enters town from the west via Italy. CR-35 runs diagonally through the western town area, terminating as it reaches the northern Hamlet of Friend.
